The Lei Lu Lab is seeking a curiosity-driven and motivated Research Fellow to investigate the dynamic organization and function of the mammalian Golgi apparatus throughout the cell cycle.

Structurally, the Golgi apparatus consists of a polarized stack of flattened membrane sacs, known as cisternae, with the cis-side functioning as the entry point for secretory cargos and the trans-side as the exit site. During secretory transport, proteins and lipids traverse the Golgi stack by sequentially moving from cis- to trans-cisternae, undergoing post-translational modifications by resident glycosyltransferases along the way. Despite its critical role in cellular function, our understanding of the Golgi's organization and trafficking mechanisms remains limited, primarily due to the technical challenges of imaging this highly dynamic and complex organelle.

Our lab has developed various quantitative and super-resolution imaging tools specifically designed for studying the Golgi, including en face averaging, side-averaging, and GLIM. These tools provide new opportunities to explore the Golgi from previously inaccessible angles. Using them, we have gathered preliminary data suggesting dynamic changes in Golgi structure and trafficking functions during the cell cycle.

As a member of our team, the successful candidate will use cutting-edge light microscopy to investigate further the molecular and cellular dynamics of the Golgi across different stages of the cell cycle. This research will contribute fundamentally to our understanding of Golgi biology and membrane trafficking.

Key Responsibilities:
  • Conduct imaging-based studies of the Golgi using advanced light microscopy techniques (e.g., spinning disk super-resolution, 3D-SIM, STED).
  • Collaborate with electron microscopy (EM) specialists to complement light microscopy with ultrastructural analysis.
  • Manage, process, and analyze large imaging datasets generated from the project.
  • Assist in mentoring and training PhD and undergraduate students in relevant techniques and concepts.

Job Requirements:
  • PhD degree in molecular and cell biology or related field.
  • At least 4 years of relevant experiences in molecular and cell biology.
  • Familiarity with confocal and wide-field light microscopy.
  • Good written and oral communication skills.
  • Proficiency in cell culture, cell imaging, and image analysis.
  • Competent in meeting presentation and report writing.
  • Interpersonal skills (Ability to work independently / develop solutions under strict timelines, meticulous and eye for details / excellent organizational / time management skills)
